چکیده

A method is described that allows the measurement of river periphyton chlorophyll a and pheophytin a both on rocks and in soft sediments. The method involves successive extractions directly from frozen samples with boiling methanol. Pigments are estimated by optical density measurements before and after acidification. Methanol is superior to acetone for the distinction of chlorophyll a and pheophytin a by this means. Estimating the mean abundance of river periphyton poses considerable problems. Because of the patchy distribution of substrates and the variability of periphyton abundance even on a single type of substrate, many samples must be examined for a good estimate of biomass. Extraction and measurement of chlorophyll are more convenient than cell counting and possibly 1 The work on which this publication is based allow a direct estimation of photosynthetic was supported in part by funds provided by the United States Department of the Interior as authopotential. While artificial substrates have rized under the Water Resources Research Act of often been used for both cell counts (see 1964, as amended. Hynes 1970; Sl&deZekovh 1962) and chloro-

Photometric method for the quantification of chlorophylls and their derivatives in complex mixtures: fitting with Gauss-peak spectra.

Accurate quantification of pigments in mixtures is essential in all cases in which separation of pigments by chromatography is impracticable for one reason or another. An example is the analysis of in vivo formation of heavy metal-substituted chlorophylls in heavy metal-stressed plants. We describe here a novel, accurate UV/VIS spectrophotometric method for the quantification of individual chlo...

Responses of marine benthic microalgae to elevated CO2

Increasing anthropogenic CO2 emissions to the atmosphere are causing a rise in pCO2 concentrations in the ocean surface and lowering pH. To predict the effects of these changes, we need to improve our understanding of the responses of marine primary producers since these drive biogeochemical cycles and profoundly affect the structure and function of benthic habitats. The effects of increasing C...

Photosynthetic performance of surface-associated algae below sea ice as measured with a pulse-amplitude-modulated (PAM) fluorometer and O2 microsensors

The photosynthetic performance of sea ice microalgae, benthic microalgae, coralline red algae (Phymatolithon foecundum) and brown macroalgae (Laminaria saccharina, Fucus evanescens and Desmarestia aculeata) in an ice-covered high-Arctic fjord (Young Sound, NE Greenland) was studied with in situ instruments for oxygen microprofiling and active chlorophyll fluorescence measurements. In situ measu...

Spectrophotometric determination of Dalacin phosphate from formulations using reduce atomic absorption of calcium ion by phosphate

A simple, sensitive and accurate method for spectrophotometric determination of Dalacinphosphatehas been established. The method is based on the reduction of atomic absorptioncalcium ion by phosphate. Because, there is a phosphate mole in every Dalacin mole structureand with measurement of phosphate, Dalacin is beyond measurement too. Therefore, the reactionbetween acidic hydrolysis products of...

SPECTROPHOTOMETRIC REACTION RATE METHOD FOR THE DETERMINATION OF NITRITE BY CATALYTIC ACTION ON NILE BLUE A BROMATE REACTION

A kinetic method developed for the determination of ultra-trace amounts of nitrite based on its catalytic effect on the reaction between potassium bromate and Nile blue A in strongly acidic media is reported. The reaction was monitored spectrophotometrically by measuring the decrease in absorbance of Nile blue A at 645 nm by the fixed-time method. The decrease in absorbance during the first...
